MOTS-c is a 16-amino acid mitochondrial-derived peptide encoded in the 12S rRNA gene of mitochondrial DNA. Discovered in 2015 by Dr. Changhan David Lee at USC, it acts as an exercise mimetic by activating the AMPK pathway, regulating the folate-methionine cycle, and promoting glucose uptake, fat oxidation, and mitochondrial biogenesis. As a key retrograde signaling molecule from mitochondria to the nucleus, MOTS-c improves insulin sensitivity, enhances physical endurance, and helps reverse age-related metabolic decline. Research shows its endogenous levels decrease with age, and supplementation demonstrates promising effects on metabolic health, inflammation reduction, and longevity.
